The cooked starch acts as the food, which the caterpillar would eat, and the alpha amylase, which is a digestive enzyme common in saliva, is stirred with it to simulate the chewing and mixing of food and saliva which constitutes the first step in the digestive system. Next two pieces of dialyses tubing were prepared by clamping one end of each piece of tubing shut with a clamp, and twisting the other end until it opens. For the purpose of the experiment, the tubing will represent the intestine of the caterpillar.Continuing on, a clean pipette was used to transfer four pipettes worth of the starch and alpha amylase solution into one of the pieces of tubing, while four pipettes of plain starch was added to the other in order to form a control. Next, two large beakers were filled two thirds of the way with distilled water. Four droppers worth of Lugol’s reagent was added two each beaker.
in nature, the bacteria can transfer these plasmids from bacteria to bacteria, transfering those beneficial genes. this mechanism allows bacteria to adapt to new and foreign environments. the continuous resistance of bacteria to antibiotics is due to the transmission of these plasmids. Bio-Rads unique pGLO plasmid encodes the gene for GFP and a gene for resistance the antibiotic ampicillin. pGLO can also control the gene expression of this fluorecent protein.
3. Why is it necessary to make pure subcultures of organisms grown from clinical specimens? So that the organism can be identified and tested for antibiotic sensitivities. 4. What kinds of clinical specimens may yield a mixed flora in bacterial cultures?

Growth media refers to any liquid or gel that will support any microorganisms or cells to grow. In the BIOE160 class we are currently using the medium LB (Lysogeny Broth or Luria Broth) to grow E. coli (this type of bacteria is usually found in the lower intestine of warm blooded organisms). Microorganisms also need water, oxygen, an energy source, carbon source, nitrogen source, and micronutrients. There are four stations that you could see on a graph of the cell growth. The first phase is called the lag phase.
