Brush biopsy of oral smears was obtained from patients with clinically normal appearing mucosa and from the lesion. The participants were informed about the nature of the study, its importance and written consent for the same in language understood by the participant was obtained. The study was conducted only after the approval from Ethics Committee of Sumandeep Vidyapeeth. The case history of participant was recorded in the prescribed format. Patients were asked to rinse the oral cavity with normal saline. Oral exfoliative smear was obtained from the buccal mucosa of controls and lesion site of study group patients. Cytosmears were obtained using a gentle scraping motion exerting little pressure with a cytobrush. Preparation of smears: • For the preparation of smears clean, fresh and dry glass slides were used. • The smears were then stained with Papanicolaou stain Orange G6 (OG-6) stain and Eosin Azure 50 (EA-50) stain. Procedure of staining: Bancroft citation29 • The slides were placed in 80% isopropyl alcohol for 2 changes of 15 minutes each to remove coating of carbowax. • Staining was performed with Papanicolaou stain Orange G6 (OG-6) stain and Eosin Azure 50 (EA-50) stain for same smear slide. • Slides were rinsed in 70% and 95% alcohol. • Slides were placed in OG-6 for 1.5 minutes. • Slides were rinsed in 95% alcohol twice. • Slides were placed in EA-50 for 1.5 to 2 minutes, then rinsed in 95% alcohol twice and absolute alcohol twice and then were cleared in xylene. • Both the slides were then mounted with DPX. • All the stained smears were observed under a light microscope. • Images were then uploaded for computer analysis.
