Pseudomonas Aeruginosa
The purpose of this lab was to identify the unknown bacteria culture given by our respected instructor using various differential tests. The identification of this unknown culture was accomplished by separating and differentiating between possible bacteria based on specific biochemical characteristics. Each test was used in a way to help recognize those specifics and bring us one step closer to identify the unknown cultures. The differential tests used to identify the unknown cultures were oxidase, lactose and glucose fermentation, citrate, indole, urease, motility, H2S, EMB and MAC.
The tests performed on the unknown bacteria culture were all used to determine and identify the bacteria. Each of the tests performed provided some key information about the bacteria in question and how it functions. Even though I performed, all the tests listed above I will only be detailing a few that I used as determinants according to my dichotomous key.
The oxidase test was used to test for the presence of cytochrome oxidase. The unknown bacteria was taken from the NA agar slant and placed on an oxidase strip in a drop of distilled water. Within seconds the bacteria will turn purple if oxidized cytochrome oxidase is present, indicating a positive test. If no color change is observed, no cytochrome oxidase is present and the test is negative. My results for oxidase test were positive. To conduct the oxidase test I first obtained an oxidase strip. I laid the oxidase strip on a paper towel. I withdrew a sample from my culture using and my inoculating loop and rubbed it on the strip. I waited for about 30 seconds and observed the strip turn purple.
